Research Focus:
Pathogens and their hosts have had a two-way chat for millions years. This interaction has led to several measure/counter-measure reactions by the host and pathogen. The host immune system has developed numerous ways to neutralize and get rid of them. In turn, pathogens have also developed mechanisms to alter and evade the host immune response. Leishmania, a protozoan parasite, clearly manipulates the host to alter the immune responses. Leishmania causes leishmaniasis which is endemic in 88 countries including India, with a total of 12 million people infected worldwide and 350 million people at risk of infection. The molecular mechanisms and consequences of host immune evasion by Leishmaniaare poorly defined. My laboratory is interested in defining these molecular mechanisms and interactions of immune evasion. I currently investigated Leishmania disrupts macrophage-membrane lipid raft and impairs p38-mitogen-activated protein kinase (p38MAPK)-mediated CD40 signaling but enhances extracellular signal-related kinase (Erk-1/2)-mediated signalling. Activation of p38MAP kinase induces pro-inflammatory cytokines like IL-12 which inhibits the growth and survival of parasite while activation of Erk-1/2 induces IL-10 production which aggravates the infection.
